Is there any risk that I might fully brick my jailbroken iPhone 4 if I perform a DFU-restore, using the same IPSW-file I used for updating it to 4.0.1?

you *cannot* brick an iPhone, *ever*. Unless it's a hardware default

DFU is part of the hardware, so software can never corrupt accessing DFU mode.
